It serves as the headquarters for the Department of Housing Preservation and Development, which is the largest municipal developer of affordable housing in the U.S. The building also houses a Senior Citizen Center for residents in lower Manhattan, the Lower East Side, and Chinatown.

Following the World Trade Center attack, 100 Gold Street served as a rest and relief center for rescue workers. Currently, plans are underway to replace this nine-story office building with a proposed 2,000-unit residential skyscraper as part of Mayor Eric Adams' "Manhattan Plan" to increase housing units in the borough.
