Sixth Avenue was originally laid out in the Commissioners' Plan of 1811, extending from Greenwich Village north to Harlem. This elevated railway was eventually demolished as part of plans to revitalize the avenue.

Starting in the 1920s, Sixth Avenue was widened and extended southward in conjunction with the construction of the Holland Tunnel and the Independent Subway System (IND) Eighth Avenue Line. This extension required the demolition of numerous buildings and the displacement of thousands of residents, primarily Italian immigrants. The redevelopment created a broader thoroughfare that allowed for the construction of subway infrastructure and new high-rise commercial buildings.

In 1945, the avenue was officially renamed "Avenue of the Americas" by Mayor Fiorello La Guardia to honor Pan-American ideals and encourage Latin American countries to open consulates along it. Despite the official renaming, New Yorkers often continued to refer to it as Sixth Avenue.

During the 1930s and beyond, the avenue saw the rise of major commercial and office buildings, including those forming Rockefeller Center, which contributed to the avenue’s prominence as a commercial and corporate corridor.
